The 2021 European Rugby Challenge Cup was the 6th season of the second tier club rugby competition in Europe. It replaced the Amlin Cup. There were 15 teams playing in 3 pools of 5 teams each. Teams played each of the other teams in their pool once. The top 3 teams from each pool and the best 4th placed team will then combine with 6 teams from the European Rugby Champions Cup to play a round of 16. This will be a knock-out format down to a finals series. |

The match between Pau and Saracens was cancelled due to positive COVID-19 tests in the Saracens team. Pau were awarded a 28-0 bonus point win.
The Worcester Warriors v Biarritz Olympique and London Irish v Brive matches were postponed due to new travel measures introduced between the UK and France. Matches were recorded as a 0-0 draw and each team received 2 points.
The match between Toulon and Newcastle Falcons was cancelled due to positive COVID-19 tests in the Newcastle team. Toulon were awarded a 28-0 bonus point win.
